SMAD5 is a member of the Mothers Against Dpp (MAD)-related family of proteins. It is a receptor-regulated SMAD (R-SMAD), and acts as an intracellular signal transducer for the transforming growth factor beta superfamily. SMAD5 is activated through serine phosphorylation by BMP (bone morphogenetic proteins) type 1 receptor kinase. It is cytoplasmic in the absence of its ligand and migrates into the nucleus upon phosphorylation and complex formation with SMAD4. Here the SMAD5/SMAD4 complex stimulates the transcription of target genes. Applications: Suitable for use in Flow Cytometry, Western Blot and Immunocytochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:100 Western Blot: 1:1000 Immunocytochemistry: 1:75 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
